I sent off my previous sample for sequencing on the previous Friday, meaning they should be returned by Monday. This did not happen, however, as sequencing data was heavily backlogged. The standard protocol is to make sure the the first mutation is correct before beginning work on the second mutation, however the lack of the sequencing data would waste the limited time I have. I decided to continue and complete the second mutation this week. At the end of the week I sent this sample off for sequencing in the hope that both mutations would be correct.
Despite this process taking slightly longer than expected, this has made me very comfortable with many molecular procedures such as PCR, colony screen and site-directed mutagenesis. This is an important learning experience that i feel will serve me well in my future research. As I will be completing a MRes in the Dale Lab, having this experience will mean i can begin research immediately without any learning period. This will allow me to make the most progress possible and ultimately get the most out of my research. Considering my research is completed in the context of learning about CMTX, making the most progress possible is a main motivating factor for me.
